Fc receptor testing is a diagnostic procedure aimed at identifying specific proteins known as Fc receptors, which play a crucial role in the immune system's defense mechanisms. These receptors are located on the surface of various immune cells, including natural killer cells, macrophages, neutrophils, and mast cells. The primary function of Fc receptors is to bind to antibodies that are attached to infecting cells or pathogens, thereby facilitating the immune response. This binding action triggers processes such as phagocytosis or cytotoxic destruction of microbes, which are essential for maintaining the body's defense against infections. There are three distinct types of Fc receptors: Fc-gamma, Fc-alpha, and Fc-epsilon. Fc-gamma receptors belong to the immunoglobulin superfamily and are instrumental in inducing the phagocytosis of microbes that have been opsonized, or marked, by antibodies. Fc-alpha receptors are present on various white blood cells, including neutrophils, eosinophils, monocytes, certain macrophages (such as Kupffer cells), and dendritic cells, contributing to the immune response. Fc-epsilon receptors, which have both high and low affinity variants, are found on cells such as epidermal Langerhans cells, eosinophils, mast cells, and basophils. The high-affinity Fc-epsilon receptors are particularly active during allergic responses and are also present on antigen-presenting cells that produce cytokines, while the low-affinity variants regulate B cell growth and inhibit IgE binding on eosinophils, monocytes, and basophils. Overall, Fc receptor testing is vital for understanding immune function and diagnosing various immunological conditions.
